Preencher ou cobrir algo completamente para que nada passe; fazer com que um buraco, tubo ou passagem fique entupido.

'Block up' é um phrasal verb, normalmente informal, usado para obstruir fisicamente (cano, buraco, porta). Frequente com 'with' (ex: block up with leaves). Mais forte que apenas 'block'. Não usado para pessoas nem contextos digitais.
