「messiest」は「messy」の最上級で、ほかの中で一番散らかったり汚かったりすることを表します。

「messiest」は部屋や机、人の性格や状態など、三つ以上のものを比べるときに使います。「the messiest room」などがよく使われます。フォーマルな場ではあまり用いません。
